ABSTRACT

The objective of this study was to evaluate egg production and quality after including crude corn oil with high acidity (CCOHA) in the feed of laying hens. The design was completely randomized to three treatments and six replicates each. The sample included 90 hens of the Lohmann LSL lineage, housed in cages; each cage contained five animals and constituted an experimental unit. The evaluated feeds were corn-based feed with degummed soybean oil (CSO), corn-based feed with CCOHA (CC) and sorghum-based feed with CCOHA (SC). At the end of the 28-day cycle, the following productivity parameters were evaluated: egg weight at laying (EWL), laying percentage (LP), feed conversion per dozen eggs (FCDZ) and feed conversion per egg mass (FCEM). The evaluated internal and external quality parameters of the eggs were yolk percentage (YP), yolk index (YI), and yolk pH (YpH); albumen percentage (AP), albumen index (AI), and albumen pH (ApH); percentage shell (PS) and shell surface area (SSA); Haugh unit (HU) and egg yolk color (EYC). There were no significant differences in EW, LP, FCDZ, and FCEM between the experimental feeds. Moreover, the use of crude corn oil with high acidity in laying hens rations did not influence the parameters of YP, YI, YpH, AP, AI, ApH, PS, SSA, HU, and EYC.


Objetivou-se avaliar a inclusão do óleo bruto de milho com alta acidez (OBMAA) em rações de galinhas poedeiras sobre a produção de ovos e sua qualidade interna e externa. O delineamento foi inteiramente casualizado com três tratamentos e seis repetições cada, com 90 galinhas da linhagem Lohmann LSL®, alojadas em número de cinco aves por gaiola, constituindo cada gaiola uma unidade experimental. Os tratamentos foram: rações base milho com óleo degomado de soja (MOS), milho com OBMAA (MOM) e sorgo com OBMAA (SOM). Ao final do ciclo de 28 dias determinou-se os parâmetros de produtividade: peso dos ovos na postura (POP), porcentagem de postura (POS) e conversões alimentares por dúzia (CAD) e por massa de ovos (CAM). Em seguida foram realizadas as análises de qualidade interna e externa dos ovos: percentagem, índice e pH da gema (GE%, IGE, pHGE), percentagem, índice e pH do albúmen (AL%, IAL, pHAL), percentagem e superfície de área de casca (CAS%, SACAS), unidade Haugh (UH) e colorimetria da gema. Não houve diferença para POS e POP, CAD e CAM das rações experimentais. A utilização do óleo bruto de milho com alta acidez nas rações de galinhas poedeiras não influenciou nos parâmetros de GE%, IGE, pHGE, AL%, IAL, pHAL, CAS%, SACAS, UH e colorimetria da gema

ABSTRACT

Biossurfactantes são compostos com características tensoativas, produzidos por diversas espécies de microrganismos. Nesse estudo, a composição de ácidos graxos de biossurfactantes produzidos por fungos filamentosos endofíticos foram analisados. Para a produção dos biossurfactantes, empregou-se fontes de carbono (óleo vegetal de milho) e nitrogênio (ureia) de baixo custo no meio de cultivo. A composição de ácidos graxos foi determinada a partir da cromatografia gasosa. Foram identificados seis picos de ésteres metílicos de ácidos graxos, com predominância do ácido linoleico (C18:2n-6c) e ácido oleico (C18:1n-9c). Tanto os ácidos graxos saturados quanto os insaturados foram encontrados na estrutura dos biossurfactantes. Estes resultados demonstraram a importância destes compostos para potenciais aplicações em várias áreas industriais.

ABSTRACT

BACKGROUND: The molecular pathways of how endocrine disruptors affect bone mineral density (BMD) and bone remodeling are still unclear. The purpose of this experimental study is to determine the effects of di(2-ethylhexyl)phthalate (DEHP) on bone metabolism in ovariectomized mice. METHODS: Twenty-six-month-old female CD-1 mice were divided into 4 groups: control, low-dose DEHP, high-dose DEHP, and estrogen groups (n=5, each group). All mice were subjected to ovariectomy for the induction of artificial menopause and then exposed to corn oil, DEHP, and estrogen for 2 months. Micro-computed tomography (Micro-CT) of the bone and analysis of blood samples for bone markers were performed to observe the changes in bone metabolism. RESULTS: Osteocalcin level was decreased in the control, low-dose and high-dose DEHP group, the reduction width was greater in the high-dose DEHP group (−0.219 ng/mL) than control group (−0.077 ng/mL, P<0.05). C-terminal telopeptide of type I collagen level was increased in the control, low-dose and high-dose DEHP group, the increase range of low-dose DEHP group (0.329 ng/mL) showed greater than control group (0.093 ng/mL, P<0.05). Micro-CT analysis revealed that the BMD was significantly lower in the high-dose DEHP group (19.8×10⁻² g/cm³) than control group (27.2×10⁻² g/cm³, P<0.05). The structure model index was significantly higher in the high-dose DEHP group (2.737) than low-dose DEHP group (2.648) and estrogen group (2.63, P<0.05). It means the progression of osteoporosis in the high-dose DEHP group. CONCLUSIONS: These results confirm the negative effects of DEHP on bone health in ovariectomized mice. Further continuous studies on genetic pathways and other endocrine disruptors will be necessary to validate these findings.

ABSTRACT

OBJECTIVE: Diabetes mellitus (DM) is known to cause many systemic complications as well as male infertility. Astaxanthin (ASTX) is a powerful antioxidant that is involved in a variety of biologically active processes, including those with anti-diabetes effects. The present study investigates the effect of ASTX on the spermatozoa function in streptozotocin (STZ)-induced diabetic rats. METHODS: We divided 30 adult rats into three groups (10 rats per group), with a control group that received corn oil mixed with chow. DM was induced by intra-peritoneal injection of STZ. Eight weeks after the STZ injection, half of the diabetic animals were used as diabetic controls, and the rest were treated with ASTX for 56 days. Then the parameters and chromatin integrity of the epididymal sperm were analyzed using chromomycin A3, toluidine blue (TB), and acridine orange (AO) staining. RESULTS: The count, viability, and motility of the epididymal sperm were decreased significantly in the STZ group in comparison with the control group (count and viability, p<0.001; motility, p<0.001;0.01). ASTX increased normal morphology and viable spermatozoa compared to the STZ group (morphology, p=0.001; viability, p<0.001;0.05). The percentage of abnormal chromatins in TB and AO staining was higher in the STZ group compared to the control group (p<0.001;0.001). The mean percentage of TB and AO positive spermatozoa in STZ rats was significantly lower in the STZ+ASTX group (TB, p=0.001; AO, p<0.001;0.05). CONCLUSION: This study observed that in vivo ASTX treatment partially attenuates some detrimental effect of diabetes. Conversely, ASTX improved sperm viability, normal morphology, and DNA integrity.

ABSTRACT

The frying by immersion is a widely used cooking process and it improves the food texture and flavor. This study analyzed the initial thermal oxidation in five edible vegetable oils used for frying. Oils samples were heated twice for 30 minutes, at 180 C and then at 240 C simulating the domestic stoves temperatures. The oils decomposition temperatures were determined by TG, being all of them > 250 C. The FA profile was analyzed by GC-FID and a slight decrease of UFA was found in corn and soybean oils. In canola, olive and sunflower oils, UFA was stable after heating treatment. Minor FA decomposition was found in canola oil, and followed by corn, olive, sunflower and soybean oils. NIR spectroscopy analyzes resulted in an extensive bands overlapping. The spectra were modeled by PCA and the oils were classified into two groups: fresh oil and heated oil, mainly by differing in 1900 nm region, associated with the carboxyl signal decrease, which might be related to the initial FA degradation in samples. It could partially understand what occurs to the vegetable oil in the beginning of its thermo-decomposition. These information are useful to consumers, food industry and health surveillance agency.


A fritura por imersão é um processo de cocção utilizado pela inclusão de textura e sabor aos alimentos. Foi analisada a oxidação térmica inicial de cinco óleos vegetais comestíveis utilizados para fritura de imersão. Amostras de óleos foram aquecidas duas vezes por 30 minutos, a 180 ºC e depois a 240 ºC, simulando-se as temperaturas de fogões domésticos. As temperaturas de decomposição dos óleos foram determinadas por TG, sendo > 250 ºC. O perfil de FA foi analisado por GC-FID, detectando-se pequeno decréscimo dos UFA nos óleos de milho e soja. Nos óleos de canola, oliva e girassol, os UFA foram estáveis após o tratamento térmico. A menor decomposição dos FA foi detectado no óleo de canola, seguido de milho, oliva, soja e girassol. Análises por espectroscopia NIR resultaram em grande sobreposição das bandas. Os espectros foram modelados por PCA, classificando-se os óleos em dois grupos: óleo fresco e óleo aquecido, principalmente pelas diferenças na região de 1900 nm, relacionadas ao decréscimo do sinal de carboxilas, e associadas à degradação inicial dos FA nas amostras. Ainda que parcialmente, pode-se entender o que ocorre com os óleos vegetais no início de termo-decomposição, abrangências que são úteis para consumidores, indústria alimentícia e órgão de vigilância sanitária.

Este estudo analisou a oxidação térmica inicial em cinco óleos vegetais comestíveis usados ​​para fritar. As amostras de óleo foram aquecidas duas vezes durante 30 minutos, a 180 ° C e depois a 240 ° C simulando as temperaturas dos fogões domésticos. As temperaturas de decomposição dos óleos foram determinadas por TG, sendo todas elas> 250 ° C. O perfil de FA foi analisado por GC-FID e uma ligeira diminuição de UFA foi encontrada em óleos de milho e de soja. ABSTRACT

Anti-atherosclerosis effects of perilla oil were investigated, in comparison with lovastatin, in rabbits fed a high-cholesterol diet (HCD). Hypercholesterolemia was induced in rabbits by feeding the HCD containing 0.5% cholesterol and 1% corn oil, and perilla oil (0.1 or 0.3%) was added to the diet containing 0.5% cholesterol for 10 weeks. HCD greatly increased blood total cholesterol and low-density lipoproteins, and caused thick atheromatous plaques, covering 74% of the aortic wall. Hyper-cholesterolemia also induced lipid accumulation in the liver and kidneys, leading to lipid peroxidation. Perilla oil not only attenuated hypercholesterolemia and atheroma formation, but also reduced fat accumulation and lipid peroxidation in hepatic and renal tissues. The results indicate that perilla oil prevents atherosclerosis and fatty liver by controlling lipid metabolism, and that it could be the first choice oil to improve diet-induced metabolic syndrome.

PURPOSE: To investigate the copaiba oil on the hepatic damage induced by acetaminophen, comparing against corn oil. METHODS: Fifty four rats were distributed into nine study groups (N=6): control group, that didn't receive the acetaminophen; Acetaminophen Group, that only received the acetaminophen; Prophylactic Copaiba Group 1, that received copaiba oil two hours before the acetaminophen; Prophylactic Copaiba Group 7, that received copaiba oil seven days, once by day, before the acetaminophen; Therapy Copaiba Group, that received the copaiba oil two hours after the acetaminophen, the corn's groups were similar than copaiba oil groups; and N-Acetyl-Cysteine Group, that received the N-Acetyl-Cysteine two hours after the acetaminophen. Euthanasia was performed after 24 hours. The serum levels transaminases, bilirubin and canalicular enzymes were analyzed. RESULTS: The prophylactic copaiba group 7, therapy copaiba group and N-Acetyl-Cysteine Group showed amounts of AST and ALT similar to the control group; and the prophylactic copaiba group 1 and corn's groups showed similar levels to the acetaminophen group. There was no significant difference between the groups regarding the amount of alkaline phosphatase and ɤ GT (p>0.05). The therapy copaiba group showed the highest levels of total bilirubin and was statistically different from the other groups (p<0.01). CONCLUSIONS: Copaiba oil administered prophylactically for seven days and therapeutically 2 hours after the acetaminophen acute intoxication offered a potential hepato protection against paracetamol-induced hepatic damage, normalizing the biochemical parameters similarly to N-Acetyl-Cysteine, and the treatment with corn oil shows no effect on the liver damage. .

ABSTRACT

Objective of this study to determine the clinical presentation and frequency of asthma triggers in the children suffering from asthma at Liaquat university of medical and health science. This cross sectional study was carried out at Liaquat University Hospital Hyderabad. All the patients were selected from OPD and pediatric ward of LUMHS. Complete history of allergy was taken from the cases and their parents and all the triggers of asthma were documented on the proforma. Total 100 patients were included in the study. Majority of the cases in the age group of 1- 3years and 2nd most common age group was 4- 7 years with the percentage of 39% and 33% respectively. Majority of the cases 51%, while moderate was present in 35% cases and 14% children were seen with severe asthma condition. Asthma triggers in the children was found as; Smoking, Exercise, Dust, Pets, Ice, Plants, Carpets, Eggs, Corn oil, Allergic rhinitis and Without triggers, with the percentage of 22.%, 55.%, 51.%, 10.%, 34.%, 05.% ,11.%, 12.% , 27.%, 46.% and17.% respectively. In the conclusion of this study we concluded that allergic triggers perform the important role in the severity of asthma disease, these triggers attacks at night and early morning, so to avoid from the smoke, dust, allergens and proper management of asthma with good compliance and regular follow up of the patients

ABSTRACT

Effects of diallyl sulfide (DAS) on thioacetamide-induced hepatotoxicity and immunotoxicity were investigated. When male Sprague-Dawley rats were treated orally with 100, 200 and 400 mg/kg of DAS in corn oil for three consecutive days, the activity of cytochrome P450 (CYP) 2E1-selective p-nitrophenol hydroxylase was dose-dependently suppressed. In addition, the activities of CYP 2B-selective benzyloxyresorufin O-debenzylase and pentoxyresorufin O-depentylase were significantly induced by the treatment with DAS. Western immunoblotting analyses also indicated the suppression of CYP 2E1 protein and/or the induction of CYP 2B protein by DAS. To investigate a possible role of metabolic activation by CYP enzymes in thioacetamide-induced hepatotoxicity, rats were pre-treated with 400 mg/kg of DAS for 3 days, followed by a single intraperitoneal treatment with 100 and 200 mg/kg of thioacetamide in saline for 24 hr. The activities of serum alanine aminotransferase and aspartate aminotransferase significantly elevated by thioacetamide were protected in DAS-pretreated animals. Likewise, the suppressed antibody response to sheep erythrocytes by thioacetamide was protected by DAS pretreatment in female BALB/c mice. Taken together, our present results indicated that thioacetamide might be activated to its toxic metabolite(s) by CYP 2E1, not by CYP 2B, in rats and mice.

ABSTRACT

This study evaluated the effect of fish oil rich in n-3 fatty acids on bone characteristics in Sprague-Dawley rats. Weanling male rats were randomized to receive either a diet containing high fish oil (FO), fish oil blended with corn oil (FICO), or soy oil rich in n-6 fatty acids (SO) for 4 weeks. All diets provided 70 g/kg fat based on the AIN-93G diet. Growth and biomarkers of bone metabolism were analyzed, and femur bone characteristics were measured by dual-energy X-ray absorptiometry. After the dietary treatment, no significant differences among the diet groups were observed for serum concentrations of Ca, parathyroid hormone, calcitonin, or osteocalcin. Alkaline phosphatase activity was significantly greater in FO-fed rats compared to that in the FICO and SO groups, whereas no difference in deoxypyridinoline values was observed, supporting the positive effect of a FO diet on bone formation. These results were accompanied by a significant increase in femur bone mineral density (BMD) in FO-fed rats. These findings suggest that providing fish oil rich in n-3 fatty acids correlates with higher alkaline phosphatase activity and BMD values, favoring bone formation in growing rats.

ABSTRACT

La situación de sobrepeso y de obesidad en la población se considera un problema grave de salud pública. Actualmente, en el mundo existen cerca de 1.600 millones de adultos con sobrepeso, de los cuales, 400 millones son clínicamente obesos. En el caso de Colombia, según la Encuesta Nacional de la Situación Nutricional, realizada en 2005 por el Instituto Colombiano de Bienestar Familiar, existe sobrepeso y obesidad en 46% de la población adulta, lo que afecta en forma más importante a las mujeres.La globalización se encuentra ligada a los rápidos cambios socioeconómicos, demográficos y tecnológicos; esto forma parte de la evolución y genera el proceso de la transición nutricional. Los seres humanos han sufrido cambios importantes en su actividad física y su estado nutricional, lo cual ha influido en el desarrollo de las enfermedades no transmisibles relacionadas con la nutrición: la urbanización, el crecimiento económico, los cambios tecnológicos en el trabajo, el procesamiento de alimentos, la cultura y el crecimiento de la información masiva.Los efectos de la modificación de los patrones implican un incremento en el consumo de alimentos, en su contenido calórico, en la disminución del gasto energético, o en todas las anteriores.Diversos estudios han correlacionado la presencia de ciertos tipos de cáncer con obesidad y la dieta es un factor muy importante para prevenirlo.Finalmente, debe considerarse la obesidad como un problema económico derivado de factores tales como políticas gubernamentales, influencia de importantes compañías de alimentos y de la inseguridad alimentaria presente en las clases socioeconómicas menos favorecidas. Es absolutamente necesario implementar programas educativos a diferentes niveles y, sobre todo, a multiplicadores de información, sobre lo que significa una dieta balanceada y la importancia de una alimentación sana en la salud y el bienestar del individuo.


Nowaday's, obesity and overweight are considered an important area in public health. Worldwide, it is estimated that nearly 1.6 billion adults are considered overweight, among which 400 million are clinically obese.The infant population is not exempt from this problem, considering that 20 million children under five are overweight. The World Health Organization (WHO) has estimated that by the year 2015 approximately 2.3 billion adults will be considered overweight and 700 million will become obese. In Colombia, according to the “Encuesta Nacional de la Situación Nutricional” conducted by the Instituto Colombiano de Bienestar Familiar (ICBF) in 2005, 46 % of the adult population appeared overweight or obese. Women, in particular, are the largest group suffering from the consequences of overweight and obesity.Globalization is linked to rapid socio-economic, demographic and technological changes, and when coupled with our natural human evolution, it generates a process of nutritional transition. Humans have had major changes in physical activity and nutritional status, which have influenced the development of non-communicable diseases related to nutrition, urbanization, economic growth, technological changes at work, food processing, culture, and the mass media messages. The changes generated in view of this inevitable interaction imply an increase in the consumption of foods leading to an increase in caloric consumption coupled with a decrease in energy expenditure. The major underlying processes that affect the world are related to the products of globalization, the information provided by the media and the influence of large multinational corporations. Soft and/or sugary drinks are important sources of caloric consumption, which stimulate factors related to obesity and increased cardiovascular risk. Barker has introduced the “fetal origin hypothesis”, which suggests that the disturbance by the plasticity of development due to malnutrition during fetal life, infancy and early childhood, permanently change the structure and function of the body by a phenomenon called “programming”. Furthermore, this becomes a determining factor in the development of a group of diseases, including type 2 diabetes, coronary heart disease, myocardial infarction, and hypertension.Because several studies have correlated certain types of cancer with obesity, diet becomes an important factor to prevent it.Finally, obesity should be regarded as an economic problem resulting from factors such as government policies, the influence of major food companies and food uncertainty, which is present in the lower socioeconomic classes. It is necessary to implement educational programs targeted at the different socioeconomic classes and, above all, to transmit the message to “information multipliers” concerning the importance of a balanced diet and incorporating healthy foods for our personal wellbeing.

ABSTRACT

This study investigated how dietary fat affects muscle atrophy and lipid metabolism in various muscles during hindlimb immobilization in rats. Twenty-four male Sprague-Dawley rats had their left hindlimb immobilized and were divided into four groups by dietary fat content and composition. The contralateral hindlimb (control) was compared with the immobilized limb in all dietary groups. Rats (n = 6/group) were fed a 4% corn oil diet (CO), 2.6% corn oil + 1.4% fish oil diet (FO), 30% corn oil diet (HCO), or a 30% beef tallow diet (HBT)after their hind limbs were immobilized for 10 days. Data were collected for the gastrocnemius, plantaris and soleus muscles. Muscle atrophy was induced significantly after 10 days of hindlimb immobilization, resulting in significantly decreased muscle mass and total muscle protein content. The protein levels of peroxisome proliferator activated receptor delta (PPARdelta) in the plantaris, gastrocnemius, and soleus increased following hindlimb immobilization irrespective of dietary fat intake. Interestingly, the PPARdelta mRNA level in the plantaris decreased significantly in all groups and that in the FO group was lower than that in the other groups. The soleus PPARdelta mRNA level decreased significantly following hindlimb immobilization in the FO group only. Muscle carnitine palmitoyl transferase 1 (mCPT1) mRNA level was not affected by hindlimb immobilization. However, the mCPT1 mRNA level in the FO group was significantly lower in the plantaris but higher in the soleus than that in the other groups. The pyruvate dehydrogenase kinase 4 (PDK4) mRNA level in the plantaris decreased significantly, whereas that in the soleus increased significantly following hindlimb immobilization. The plantaris, but not soleus, PDK4 mRNA level was significantly higher in the FO group than that in the CO group. The increased PPARdelta protein level following hindlimb immobilization may have suppressed triglyceride accumulation in muscles and different types of dietary fat may have differentially affected muscle atrophy according to muscle type. Our results suggest that omega-3 polyunsaturated fatty acids may suppress muscle atrophy and lipid accumulation by positively affecting the expression level and activity of PPARdelta and PPARdelta-related enzymes, which are supposed to play an important role in muscle lipid metabolism.

ABSTRACT

Investigou-se o efeito da adição de C18:2n6, por meio da inclusão de óleo de milho em dietas com dois níveis de proteína bruta, sobre o processo de maturação de gametas de pintado, Pseudoplatystoma corruscans, mantidos em tanques-rede. Foram avaliados: taxa de sobrevivência, relação peso x comprimento, fator de condição (K) e índice gonadossomático (IGS). O experimento foi realizado entre março de 2004 e fevereiro de 2006, em 12 tanques-rede, distribuídos em seis viveiros-escavados de 600m² e densidade de estocagem de 20 peixes/tanque-rede. Utilizaram-se três tratamentos (T) com duas repetições/viveiro: T1 com 28 por cento de PB; T2 com 28 por cento de PB + 5 por cento óleo de milho e T3 com 40 por cento de PB. O crescimento foi ligeiramente mais alto nos peixes do T3. As taxas de sobrevivência foram acima de 77 por cento. Pode-se inferir que as rações ofertadas não causaram alterações histomorfológicas durante o processo de maturação gonadal dessa espécie. O IGS e o K foram ligeiramente mais altos nos animais alimentados com a ração enriquecida com óleo de milho.


It was studied the effect of the addition of C18:2n6, by the inclusion of corn oil, in diets with different levels of crude protein (CP) on the process of gonad maturation in surubim, Pseudoplatystoma corruscans. Survival rate, weight x length ratio, condition factor (K), and gonadossomatic index (GSI) were evaluated. The experiment was carried out from March 2004 to February 2006, using 12 cages distributed in six tanks, with 20 fishes per cage. Three treatments (T) were: T1 28 percent CP; T2 28 percent CP + 5 percent corn oils, and T3 40 percent CP. No effect of the diet was observed on growth in the 1st and 2nd years of age, with a slightly superior growth of T3 fish. The survival rates were superior to 77 percent. It can be inferred that the offered diet did not cause histomorphological alterations during the process of gonadal maturation of this species. However, the GSI and the K were slightly advanced and superior in the animals fed the diet supplemented with corn oil.

ABSTRACT

PURPOSE: Polychlorinated biphenyls (PCBs, Aroclor 1254), synthetic chlorinated organic compounds, are known to decrease thyroid function, sperm count, and fertility, and increase the risk of testicular cancer; they may have serious effects on male reproduction. The aim of this study was to investigate the possible protective role of palmiwon on PCB-induced spermiotoxicity in rats. MATERIALS AND METHODS: 90-day-old male Sprague Dawley rats were randomly divided into three groups, each consisting of ten animals. The control group (Group I) received corn oil, the second group of rats (Group II) was administered 2 mg/kg body weight/day of Aroclor 1254+corn oil intraperitoneally for 30 days. The third group of rats (Group III) was treated with 2 mg/kg body weight/day of Aroclor 1254+corn oil intraperitoneally plus palmiwon (300 mg/day) orally for 30 days. Twenty-four hours after the last treatment, the animals were killed by decapitation. Their serum testosterone levels was measured before and after the experimental medication was taken, and the number and motility of sperm, which had been collected from the cauda epididymal region, were evaluated. RESULTS: The results of this experiment show that treatment with palmiwon significantly improved sperm motility and number in rats that had been exposed to PCBs. There was no marked difference in body weight, testis weight, or epididymis weight among the groups. Nor were there any significant pathological differences in the testes among the groups. CONCLUSIONS: Palmiwon has the potential for treating PCB-induced spermiotoxicity. More experiments with larger samples and a longer period of treatment are needed.

ABSTRACT

BACKGROUND: The present study was undertaken to examine the immunological effects of pentabrominated diphenyl ether (penta-BDE) and decabrominated diphenyl ether (deca-BDE) on the immune system of the dams and the developmental immune system of the offsprings. METHODS: In this study, mated female C57BL/6J mice were orally administered penta-BDE, deca-BDE or corn oil for 5 weeks, from gestational day 6 to lactational day 21. RESULTS: The body weight of PND21 exposed to penta-BDE was significantly decreased relative to control mice, but that of post-natal day 63 (PND63) were recovered. Orally dosed dams with penta-BDE had significantly smaller absolute and relative spleen masses than control mice. Absolute and relative spleen and thymus masses of PND21 exposed to penta-BDE were significantly decreased over control. The exposure of dams and PND21 with penta-BDE reduced the number of splenocytes and thymocytes. As results of hematologic analysis, percentage WBC and percentage neutrophils increased in dams with deca-BDE. Splenic T cell proliferation in dams and PND21 exposed to penta-BDE was increased, and there were no significant difference in splenic B cell proliferation in all treatment groups. As results of flow cytometric analysis of splenocyte, percentage total T cell, Th cell and Tc cell in PND21 exposed to penta-BDE was slightly increased, and percentage macrophage in dams and PND21 exposed to deca-BDE was decreased. The ELISA results of antibody production show no significant difference in all treatment groups relative to controls. CONCLUSION: These results imply that PBDEs given to the dam were transferred to the offspring during gestation and lactation, and PBDEs transferred from the dam affect immune system of offspring.

ABSTRACT

It has been reported that dietary polyunsaturated fats (PUFA) increase liver injury in response to ethanol feeding. We tested the hypothesis that diets rich in linoleic acid (18:2n-6) would affect acute liver injury after acetaminophen injection and that protein restriction might exacerbate the liver injury. We examined effects of feeding diets with either 15% (wt/wt) corn oil or 14% beef tallow and 1% corn oil for six weeks with either 6 or 20 g/100 g protein on acute hepatotoxicity. After the feeding period, liver injury was induced by injecting either with 600 mg/kg body weight acetaminophen suspended in gum arabic-based vehicle, or with vehicle alone during fasting status. Samples of liver and plasma were taken for analyses of hepatic glutathione (GSH) levels and liver-specific enzymes [(Glutamate-pyruvate transaminase (GPT) and glutamate-oxaloacetate transaminase (GOT)], respectively. Whereas GSH level was significantly lower in only group fed 15% corn oil with 6 g/100 g protein among acetaminophen-treated groups, activities of GPT and GOT were significantly elevated in all groups except the one fed beef tallow with 20 g/100 g protein, suggesting low protein might exacerbate drug-induced hepatotoxicity. The feeding regimens changed the ratio of 18:2n-6 to oleic acid (18:1n-9) in total liver lipids approximately five-fold, and produced modest changes in arachidonic acid (20:4n-6). We conclude that diets with high 18:2n-6 promote acetaminophen-induced liver injury compared to diets with more saturated fatty acids (SFA). In addition, protein restriction appeared to exacerbate the liver injury.

ABSTRACT

PURPOSE: To test the effects of a low fat diet compared with a babassu fat diet on nutritional status in obstructive cholestasis in young rats. METHODS: We submitted 40 rats in 4 groups of 10 animals each from P21 (21st postnatal day) to P49 to two of the following treatments: bile duct ligation or sham operation and low fat diet (corn oil supplying 4.5 percent of the total amount of energy) or babassu fat diet (this fat supplying 32.7 percent and corn oil supplying 1.7 percent of the total amount of energy). Weight gain from P25 to P49 every 4 days was measured. The Verhulst's growth function was fitted to these values of weight gain. Growth velocity and acceleration at each moment were estimated using the same equation. Total food and energy intake from P21 to P49, energy utilization rate (EUR) from P25 to P49 and fat absorption rate (FAR) and nitrogen balance (NB) from P42 to P49 were measured. Two Way ANOVA and the S.N.K. test for multiple paired comparisons were employed to study the effects of cholestasis and those of the diets and their interaction (p<0.05) on those variables. RESULTS: In cholestatic animals, a higher growth velocity at P45, a higher growth acceleration at P41 and P45, a greater EUR, a greater FAR and a greater NB, were found with the low fat diet as compared with the babassu fat diet. CONCLUSION: A low fat diet lessens the growth restriction brought about by cholestasis and allows for an improved dietary energy utilization and a better protein balance than the babassu fat diet.


OBJETIVO: Testar os efeitos de uma dieta com baixo teor de gordura comparada a uma dieta com gordura de babaçu sobre o estado nutricional em ratos jovens com colestase obstrutiva. MÉTODOS: Submetemos 40 ratos divididos em quatro grupos de 10 animais a partir do P21 (21º dia pós-natal) até o P49 a dois dos seguintes tratamentos: ligadura e ressecção do ducto biliar comum ou operação simulada e dieta com baixo teor de gordura (óleo de milho fornecendo 4,5 por cento da quantidade total de calorias) ou dieta com gordura de babaçu (essa gordura fornecendo 32,7 por cento e óleo de milho fornecendo 1,7 por cento da quantidade total de calorias). Foi mensurado o ganho de peso a cada 4 dias do P25 ao P49. A função de crescimento de Verhulst foi ajustada aos valores de ganho de peso. A velocidade e a aceleração de crescimento nos mesmos momentos foram estimadas usando a mesma equação. Foram mensurados: quantidade de ração ingerida e ingestão energética total do P21 ao P49, utilização de energia do P25 ao P49, gordura absorvida e balanço de nitrogênio (BN) do P42 ao P49. A ANOVA com dois fatores e o método de S.N.K para comparações pareadas foram utilizados para estudar os efeitos, sobre as variáveis, da colestase e das dietas e sua interação (p<0,05). RESULTADOS: Em ratos com colestase e dieta com baixo teor de gordura, houve maior velocidade de crescimento no P45, maior aceleração de crescimento no P41 e P45, maior utilização de energia, maior percentual de gordura absorvida e maior BN do que em ratos com colestase e dieta com gordura de babaçu. CONCLUSÃO: A dieta com baixo teor de gordura atenua a restrição de crescimento provocada pela colestase e proporciona melhor aproveitamento da dieta e maior incorporação da proteína ingerida do que a dieta com gordura de babaçu.

ABSTRACT

Obesity has become a worldwide health problem. Orlistat, an inhibitor of pancreatic lipase, is currently approved as an anti-obesity drug. However, gastrointestinal side effects caused by Orlistat may limit its use. In this study the inhibitory activities of dandelion (Taraxacum officinale) against pancreatic lipase in vitro and in vivo were measured to determine its possible use as a natural anti-obesity agent. The inhibitory activities of the 95% ethanol extract of T. officinale and Orlistat were measured using 4-methylumbelliferyl oleate (4-MU oleate) as a substrate at concentrations of 250, 125, 100, 25, 12.5 and 4 microgram/ml. To determine pancreatic lipase inhibitory activity in vivo, mice (n=16) were orally administered with corn oil emulsion (5 ml/kg) alone or with the 95% ethanol extract of T. officinale (400 mg/kg) following an overnight fast. Plasma triglyceride levels were measured at 0, 90, 180, and 240 min after treatment and incremental areas under the response curves (AUC) were calculated. The 95% ethanol extract of T. officinale and Orlistat, inhibited, porcine pancreatic lipase activity by 86.3% and 95.7% at a concentration of 250 microg/ml, respectively. T. officinale extract showed dose-dependent inhibition with the IC50 of 78.2 microg/ml. A single oral dose of the extract significantly inhibited increases in plasma triglyceride levels at 90 and 180 min and reduced AUC of plasma triglyceride response curve (p<0.05). The results indicate that T. officinale exhibits inhibitory activities against pancreatic lipase in vitro and in vivo. Further studies to elucidate anti-obesity effects of chronic consumption of T. officinale and to identify the active components responsible for inhibitory activity against pancreatic lipase are necessary.
